In a test tube, do queens require water? I keep them with water, but I just would like to know why it's done that way. Humidity, water for drinking etc?

Oh boy.. Lots to learn, no offense.
Ants themselves like virtually all living creatures, require water to survive. Ants, depending on species, can last from 1-7 days without water.
Moisture and humidity play a huge role when it comes to brood development, particularly for eggs and larvae.
So, in short, yes you must give them a water source.

If you have a nest you can't easily get wet, and it doesn't soak up moisture, feed them sugar water. If it's a flat nest, have a tube leading to a foraging box. Inside the box have a small bowl, which you place sugar water in.

Yes but put some cotton in so they don't drown, also ants nest have to be pretty moist so if you have a Ytong nest you'll need to water that
