How do you know they are not hiding underground? Also my messors are really bad at walking over talcum power. But if you are really sure they escaped then all I can think of is placing some seeds near the nest and wait a day to see if some seeds are missing.. so only lay down a small amount.

I really doubt they have escaped as my messor queen falls when walking plastic or glass and I suppose that if the queen cant go out thw workers wouldnt go out... Are you sure they havent dug a small hole or anything?

As long as you keep wood dry and have no ants that nest in wood, it should be fine. But messor can chew trough ytong blocks, no idea if that's harder or softer then wood (depends on the type of wood I guess). Personally I would just make the nest the best place for them to be in, cover up the tube. Make sure they have water and seeds. Keep the nest warm, etc.. and then hope when one of the workers walks back for water/seeds and that she'll find the nest a better place to stay then there current location. That should mean she'll let the others know and that they'll move back.

Yes and no, you would need to keep them in one way or another. Stevant used talk powder, should normally do the trick.
